Butyryl coenzyme A dehydrogenase: studies on the specificity of the formation of acyl coenzyme A complexes with long-wavelength absorption.
نویسنده
چکیده
Further studies of butyryl CoA dehydrogenase show that the enzyme forms tight complexes with long-wavelength absorption bands upon addition of a,^-unsaturated alicyclic carboxyl ic acyl CoA compounds. A s in the straight chain series, the size of the acyl moiety is critical. The thiol portion by contrast determines neither whether a long-wavelength band is formed nor the wavelength of maximum absorption if such a band is present. It does however markedly affect the dissociation constant. Unusual features of the acid stability of the acyl C o A found on the native green form of the enzyme are also reported.

Acyl-coenzyme A synthetases.
The enzymes catalysing the initial stage of the 8-oxidation of fatty acids, the acyl-CoA synthetases, have been classified into four groups based on specificity. These are: the short-chain (acetyl-CoA synthetase; EC 6.2.1 .l), medium-chain (butyryl-CoA synthetase; EC 6.2.1.2) and the long-chain fatty acyl-CoA synthetase (acyl-CoA synthetase; EC 6.2.1.3), which are ATP-dependent and follow the r...
متن کاملSuccessful management of pregnancy with very-long-chain acyl-coenzyme A dehydrogenase deficiency.
Very-long-chain acyl-coenzyme A dehydrogenase deficiency (VLCADD) is a rare and life-threatening disease characterized by an enzymatic defect in the fatty acid β-oxidation pathway. A nulliparous woman with VLCADD showed improvements in serum levels of the long-chain acylcarnitine moiety (C14:1) during pregnancy and successfully delivered a healthy infant vaginally. Pregnancy and vaginal deliver...
متن کاملPurification and role of phosphotransbutyrylase.
In recent years acyl phosphates have received much attention. For example, acetyl (1, 2), propionyl (3, 4), butyryl (5, 6), carbamyl (7, 8), and aspartyl (9) phosphates have been studied in detail. In fermentative bacteria the role of phosphotransacetylase, which facilitates an interconversion of acetyl phosphate and acetyl coenzyme A has been well documented (1,2,10-12). An enzyme capable of c...
متن کاملGenetic deficiency of short-chain acyl-coenzyme A dehydrogenase in cultured fibroblasts from a patient with muscle carnitine deficiency and severe skeletal muscle weakness.
Genetic deficiency of short-chain acyl-coenzyme A (CoA) dehydrogenase activity was demonstrated in cultured fibroblasts from a 2-yr-old female whose early postnatal life was complicated by poor feeding, emesis, and failure to thrive. She demonstrated progressive skeletal muscle weakness and developmental delay. Her plasma total carnitine level (35 nmol/ml) was low-normal, but was esterified to ...

The 2-oxoacid dehydrogenase complexes (ODHc) consist of multiple copies of three enzyme components: E1, a 2-oxoacid decarboxylase; E2, dihydrolipoyl acyl-transferase; and E3, dihydrolipoyl dehydrogenase, that together catalyze the oxidative decarboxylation of 2-oxoacids, in the presence of thiamin diphosphate (ThDP), coenzyme A (CoA), Mg²⁺ and NAD⁺, to generate CO₂, NADH and the corresponding a...
